Mechanism of Action (MoA)
As a derivative of the nitro-dihydro-imidazooxazole class, Delamanid functions as a prodrug. Once inside the Mycobacterium tuberculosis cell, it is activated by the mycobacterial F420 coenzyme system (including the enzyme Ddn) to form a reactive metabolite.
- Target: This reactive intermediate selectively inhibits the synthesis of methoxy-mycolic acid and keto-mycolic acid, which are vital components of the mycobacterial cell wall.
- Result: This powerful targeted action disrupts the cell wall structure, leading to bacterial death in both actively growing and non-replicating (dormant) M. tuberculosis strains.
